You do need to restart MailMate for changes to take effect.
If not restarting wasn't the problem, you can check the format of the
file:
~$ plutil -lint Library/Application\
Support/MailMate/Resources/Layouts/composerHeaders.plist
Library/Application
Support/MailMate/Resources/Layouts/composerHeaders.plist: OK
